|
Ultraviolet(UV) radiation is a prevailing factor implicated in the etiology of actinic keratosis(AK) and squamous cell carcinoma(SCC), as evidenced by the high frequency of UV related mutations in the p53 and p16 tumor suppressor genes. We aimed to evaluate p53 and p16 expression in the progression of squamous cutaneous neoplasia and observe whether p53 and p16 expression varies with different etiologies of SCC in this study. P53 and p16 expression in 31 cases of SCC(7 UV-induced, 3 arsenic-induced, 3 arising in scars, 4 HPV-related, 2 immunosuppression-related, 2 de novo, 4 oral and 6 periungual), 19 cases of AK(6 low, 7 intermediate and 6 high-grade) and 11 cases for controls(3 hypertrophic scar, 5 wart and 3 normal skin) was studied by using an immunohistochemical method. P53 immunostaining was positive in 13 of 31 SCC cases, 10 of 19 AK cases and none of 11 controls. P16 was positive in 17 of 31 SCCs, 7 of 19 Aks and 1 of 11 controls. Poorly differentiated SCC showed more increased p53 and p16 expression than well-differentiated SCC statistically insignificantly. There was a higher frequency of increased p53 and p16 expression in UV-induced compared with SCC of other etiologies statistically insignificantly. In conclusion, there was a direct correlation between the increased expression of p16 during the progression from mild AK to severe AK to squamous cell carcinoma statistically significantly. P53 and p16 expression varied with different etiologies of SCC.
